His career at Second City culminated in the winter of 2016 when Perkins co-wrote and starred in "Black Side of the Moon", a collaboration between Second City and Woolly Mammoth Theatre Company. The show is known as one of Second City's best selling shows in history.

In April 2017, Perkins joined the cast of the 9th season of MTV's Wild 'n Out[5] as the first openly gay male in the history of the show.

Perkins is a part of the award winning comedy collective "3Peat," alongside Saturday Night Live cast member Chris Redd. 3Peat's sketch "The Blackening"[6], which Perkins wrote for Comedy Central, went viral in 2018. The sketch is being turned into a feature film co-written by Perkins and Tracy Oliver (Girls Trip, Little), to be produced by MRC Studios and The Story Company. Perkins is developing a solo comedy show How Being Black and Gay Made Me Better Than You. The show serves as an interdisciplinary experience showcasing some aspects of Dewayne's background in dance and theatre as he is a technically trained ballet and hip-hop dancer, while chronicling his life as a black gay man in comedy. The show previewed in Chicago in December of 2019 and the official one-man show will be going up in 2020.

As a stand up comedian Perkins has opened for Hannibal Buress and Joel Kim Booster, and performed all over the country at venues like The Comedy Store, The Laugh Factory and the New York Comedy Club, and festivals like NBC's Break Out Comedy Festival, Accidental Comedy Festival, SF Sketchfest, and Comedy Central's Clusterfest.
