The Human Vaccines Project (HVP) is a global nonprofit organization with the mission to decode the human immune system and accelerate the development of effective vaccines and immunotherapies across major global diseases.[1] [2]. Scientists at the Human Vaccines Project merge biomedicine with artificial intelligence and machine learning [3] to understand how the immune system fights illness, whether non-communicable diseases such as cancer, diabetes, and Alzheimer’s, or infectious ones such as HIV, tuberculosis, malaria [4], and newly emerging disease threats like COVID-19 [5].

The Human Vaccines Project aims to understand the rules of immunity across different population groups [6], create the first AI-model of immunity and establish a blueprint for next generation vaccines and immunotherapies [7].

The Project is modeled after the Human Genome Project [6], the moonshot initiative that transformed biomedical research, the future of drug development and personalized medicine [8] [9].

History

The foundation for the Human Vaccines Project was laid on February 6, 2014 when 35 leading vaccine scientists – amongst them current CEO Wayne C. Koff and Nobel Laureate Peter Doherty – met in La Jolla, California to discuss how to harness the technological advances in antigen discovery, genomics and immunological monitoring to revolutionize vaccine development [9]. Over the next months, the Human Vaccines Project developed a scientific plan and management structure [10] and began full-time operations in April 2016 [11].

Leadership & Notable Members

Wayne C. Koff, Ph.D. is founding president and CEO of the Human Vaccines Project. Prior to joining HVP [11], he served as chief scientific officer and senior vice president of research and development at the International AIDS Vaccine Initiative (IAVI) in New York City (1999-2016), leading IAVI’s research and development program [12]. Wayne C. Koff was honored by Foreign Policy as one of the 100 Global Thinkers in 2019 along with Barack Obama, Angela Merkel, and Bill Gates [13]. In 2020, Koff was appointed adjunct professor of epidemiology at the Harvard T.H. Chan School of Public Health [14].

Ted Schenkelberg, M.B.A., M.P.H., is Co-Founder and COO of the Human Vaccines Project. He brings more than 15 years of experience applying business management principles to advance biomedical and global health priorities across the nonprofit and for-profit sectors [15].

The Human Vaccines Project operates with a core leadership team, board of directors, scientific steering committee and futures council[16]. Notable members of the board are Julie Bishop, M.D., former Australian Minister for Foreign Affairs, Jane Metcalfe, Founder/CEO of NEO.LIFE and WIRED Co-Founder, José María Fernández, M.B.A., M.F., former director general of the Treasury at Spain’s Ministry of Economy and Competitiveness, Marie-Paule Kieny, Ph.D. as well as renowned vaccine researcher Stanley Plotkin, M.D. [17]

The scientific steering committee has included, among others, Nobel Laureate Peter Doherty, M.D., Barney Graham, M.D., Ph.D, Deputy Director of the Vaccine Research Center at NIAID at NIH, Kathrin Jansen, Senior Vice President and Head of Vaccine Research & Development at Pfizer, Andrea Carfi, Ph.D., head of research for infectious disease at Moderna, and Soumya Swaminathan, M.D., former Deputy Director-General for Programmes at the World Health Organization [18]. Members of the Human Vaccines Project futures council are Pulitzer Prize-winning author Laurie Garrett, an internationally recognized expert on global health and disease prevention, Ann Druyan, a Peabody and Emmy Award-winning writer, producer and director specializing in the communication of science, and Karl-Theodor zu Guttenberg, former German Minister of Economics and Technology [19].

Initiatives

The Human Vaccines Project established key initiatives to focus on those populations most at risk of disease and the hardest to protect:

Human Immunomics Initiative

The Human Immunomics Initiative (HII) in collaboration with Harvard T.H. Chan School of Public Health [20] aims to extend healthy life spans by determining the underlying rules of human immunity, with a focus on aging adults [21].

Newborn Immunity Initiative

The Newborn Immunity Initiative (NII) in partnership with the Australian Telethon Kids Institute has the goal to reduce infant mortality by harnessing the unique immunity of mothers and infants to improve pregnancy outcomes and reduce infant disease and death [22].

Coronavirus Vaccine Initiative

The Coronavirus Vaccine Initiative (CVI) advocates the development of a universal coronavirus vaccine, which protects against viruses across the whole coronavirus family [23]. Part of this initiative are two regular formats by the Human Vaccines Project: a bi-weekly newsletter, the HVP COVID Report, as well as a webinar, the Global COVID Lab Meeting, on alternating Thursdays [24].

Scientific Highlights

In collaboration with the Vanderbilt University Medical Center and Scripps Research, the Human Vaccines Project conducted the first sequencing of the human immunome. The study was lead by James E. Crowe and published in Nature; it showed the potential of decoding the human immune system by merging cutting-edge biological research with frontier computing advancements [25].

HVP conducted a study of human immune responses to hepatitis B vaccination, establishing the possibility to predict who will respond to vaccination by combining systems biology and machine learning [26].

In a study published in Nature in collaboration with Harvard T.H. Chan School of Public Health, HVP showed that accelerated biological age for a given age is a predictor of major age-related morbidity, including dementia, among healthy elderly [27]).

Collaborations

The Human Vaccines Project created a network of leading university and academic research centers that serve as scientific hubs and work collaboratively. HVP collaborates with governmental, academic, nonprofit, and industrial affiliates to generate ideas, share data, fund projects and advance key scientific goals [28].
