Route 349 commenced operation on 24 April 2004 between Ponders End Enfield Garage and Stoke Newington Common using Alexander ALX400 bodied DAF DB250LFs. The new route was introduced to replace route 149 between Edmonton Green and Ponders End.
On 1 April 2006, the allocation was transferred to Enfield (E) garage.
In October 2009, Plaxton President bodied DAF DB250LFs were introduced.
On 16 October 2010, the route was retained by Arriva London using Wright Eclipse Gemini bodied Volvo B7TLs. The route was withdrawn between Stamford Hill and Stoke Newington at the same time[1], this part was replaced by the revised Route 73 where it got withdrawn between Stoke Newington & Seven Sisters.[2]
In 2014, existing Wright Pulsar Gemini bodied DAF DB250LFs were introduced alongside the Wright Eclipse Gemini bodied B7TLs.
On 17 October 2015, the route was retained by Arriva London using existing Wright Gemini 2 bodied VDL DB300s from the Route 149 when the contract renewal for the Route 149 to New Routemasters.
On 9 April 2016, the route was revised to run in service from Stamford Hill to the garage in Rookwood Road.
On 16 September 2021, existing New Routemasters were introduced alongside the Wright Gemini 2 bodied VDL DB300s as a spare from Routes 38, 73, 253, 254 and 313.[3]
In November 2021, the route was converted to full New Routemaster operation.
In March 2022, the iBus system for the 349 got updated for its destination for Ponders End.

New: "349 to Ponders End Enfield Bus Garage"
The only subtle difference is adding Enfield, this was done to address Ponders End Bus Garage is in Enfield(E) as evident from bus stop B namely "Enfield Bus Garage/ Southbury Station

Previous Route 349 in London
Ex. 349 was operating in the south-west of London from West Brompton Empress State Building to Streatham Garage(Mondays to Saturdays) and Crystal Palace(Sunday) operated by South London. The route was introduced in 1991 to partly withdraw Route 49.[4]
A year later, Ex. 349 diverted at St Leonards Church to Brixton Garage, the Sunday only section was replaced by Route 249 introduction on this day and the destination for Streatham was changed to Streatham Telford Avenue.
The route was completely withdrawn in 1993 having only been operating for only three years, this route got replaced by Route 319.[5]
