Marc E. Rothenberg, MD, PhD is an American Physician-Scientist and Professor of Pediatrics. He is the Founder and Director of the Division of Allergy and Immunology at Cincinnati Children’s Hospital Medical Center (CCHMC) and a tenured Professor of Pediatrics at the University of Cincinnati College of Medicine (UCCOM). Rothenberg's research has elucidated fundamental mechanisms involved in allergies, particularly focused on eosinophils and control of their expansion. His research paved the way for the new class of asthma medicines (anti-eosinophil therapies), which are now being applied to other diseases such as hypereosinophilic syndrome and eosinophilic gastrointestinal diseases (EGID). His pioneering research has led to the recognition of EGID as a separate disease entity, with a special interest in understanding the genetic, cellular, and molecular basis for eosinophilic esophagitis (EoE).

Career

Rothenberg graduated summa cum laude with Highest Honors in Chemistry and Biochemistry from Brandeis University. His research at Brandeis University was conducted under the mentorship of Professor William P. Jencks, MD, a pioneer in biochemistry and enzymology. At Harvard Medical School (HMS), he completed the combined MD/PhD program. His PhD, under the mentorship of Professor K. Frank Austen, included seminal studies on eosinophil hematopoiesis, identifying and characterizing the original biology of eosinophilopoetins, which helped lead to the development of a new class of FDA drugs (humanized anti–IL-5). Dr. Rothenberg completed a pediatric residency and combined fellowship in Allergy/Immunology and Hematology at Children’s Hospital in Boston. His postdoctoral training was completed in the genetics laboratory of Professor Philip Leder, MD at HMS, where he cloned the eotaxin chemokine, resulting in a new direction in allergy research. He came to the University of Cincinnati College of Medicine and Cincinnati Children's Hospital Medical Center in 1996 and has helped build a renowned program in pediatric research.

In 2001, Rothenberg established the Cincinnati Center for Eosinophilic Disorders (CCED), the first center solely focused on eosinophilic diseases. The CCED serves as a model and is exemplary to other centers formed around the world. Rothenberg also founded and is the current Director of the NIH-sponsored Consortium of Eosinophilic Gastrointestinal Disease Researchers (CEGIR) involving 16 clinical and research centers in the USA and part of the Rare Diseases Clinical Research Network (RDCRN), led by the National Center for Advancing Translational Sciences [NCATS] through its Office of Rare Diseases Research [ORDR] of the NIH. CEGIR’s mission is to improve the lives of those suffering from eosinophilic gastrointestinal disorders. EGIDExpress is the data-sharing platform for researchers to view published datasets generated by the Rothenberg CURED Laboratory.

Research

Rothenberg leads the Rothenberg CURED Laboratory at Cincinnati Children's Hospital Medical Center in conducting basic, translational, and clinical research. Rothenberg's research focuses on the molecular analysis of allergic inflammation, primarily on the pathogenesis of eosinophilic gastrointestinal disorders (EGIDs). His laboratory takes multidisciplinary approaches including the development of preclinical murine models of allergic disease, genetics, genomics, molecular immunology, cellular biology, and biochemistry.

Rothenberg has elucidated the pathways that control eosinophilia, involving the eosinophilopoeitin IL-5 and the eotaxin family of chemoattractants, and propelled the development of a new class of drugs including conducting proof-of-principle studies in humans. He defined the role of T cell derived IL-13, the disrupted epithelial barrier elicited by imbalanced proteases/anti-proteases, and microRNAs in eliciting allergic esophagitis. His findings have provided the basis for molecular diagnostics and precision therapy in the allergy field such as the eosinophilic esophagitis diagnostic panel (EDP).[1]

Rothenberg is an international speaker, most often for his work on Mechanisms of Allergy and Eosinophilic Esophagitis (EoE).

Awards and Honors

Rothenberg’s awards include the 2007 E. Mead Johnson Award from the Society of Pediatric Research and an NIH MERIT Award in 2010, and he was recognized as a Highly Cited (top 1%) Researcher for 2018 (Clarivate Analytics). He is an elected member of the American Society of Clinical Investigation, American Academy of Pediatrics, American Association for the Advancement of Science, Society for Pediatric Research, and Hewlett-Woodmere Alumni Hall of Fame. Rothenberg's publications number over 400. He is an Associate Editor of the Journal of Allergy and Clinical Immunology and an Editorial Advisor for the Journal of Clinical Investigation and co-Section Head of the Allergy and Hypersensitivity Section of Faculty 1000. He has served a four-year term on the Advisory Council of the NIAID, during which the council advised NIAID Director Anthony Fauci on the allocation and prioritization of its $4 billion annual budget. Rothenberg's research has been supported by numerous sources including the NIH, Human Frontier Science Program Organization, Burroughs Wellcome Fund, Dana Foundation, [United States Department of Defense], US-Israel Binational Fund, and Patient-Centered Outcomes Research Institute. He has trained over 100 clinical and research investigators in his laboratory and also as Program Director and/or Co-Principal Investigator of several NIH training grants.
