Pseudomonas virus phi8

From EverybodyWiki Bios & Wiki

Φ8 (phi8) is a bacteriophage that belongs to family Cystoviridiae. It was isolated from leaved of pea plant (Pisum sativum)[1]. Φ8 uses mostly Pseudomonas as host. Unlike Φ6, which uses type IV pili to enter cells, Φ8 uses host lipopolysaccharides[2].

RNA dependent RNA polymerase (RdRp)

Like the RNA polymerase of Φ6, the RNA polymerase of Φ8 works without primers[3]. Despite having low sequence identity to the polymerase of Φ6, the two polymerases fold similarly. Like the Φ6 polymerase, the polymerase of Φ8 has high affinity to templates ending with cytidine. In addition to that, it favors sequences with many uridines in the beginning. The polymerase has two metal ion binding sites, one of them being in the catalytic pocket. It is assumed that Mg2+ or Mn2+ ions are bound to the sites when the polymerase is in active state, and Ca2+ when it is in inactive state.
