Stefan Rose-John
Stefan Rose-John (*December 17, 1954) is a German Biochemist and emeritus professor at the Christian-Albrechts-University of Kiel.
Education and carrier
Rose-John studied biologie with subsidiaries chemistry and physics at the University of Heidelberg. After obtaining his PhD in Heidelberg he obtained research grants from the Max-Kade Foundation and the Deutsche Forschungsgemeinschaft to work at the DOE-Plant Research Laboratory at the Michigan State University in the US. After 3 years as a group leader at the German Cancer Research Center in Heidelberg, he went to the RWTH Aachen where he obtained his habilitation in Biochemistry. 1994 he became professor of Pathophysiology at the Johannes Gutenberg-University in Mainz. In 2000 he was appointed full professor and director of the Biochemical Institute at the Medical School of the Christian-Albrechts-University of Kiel. In 2010 he founded the Collaborative Research Center 8777 (Proteolysis as a regulatory event in pathophysiology), which was financed by the Deutsche Forschungsgemeinschaft. At the end of 2022 he became an emeritus professor at the Christian-Albrechts-University of Kiel.
Field of Research
Area of expertise Rose-John works in the field ofcytokines and their activities in inflammation and cancerogenesis. He was the first to describe the Interleukin-6 trans-signaling pathway, in which the cytokine Interleukin-6 (IL-6) acts via a soluble IL-6 receptor can stimulate all cells in the body. He could show that the pro-inflammatory aktivities of IL-6 are mediated by the soluble IL-6 receptor [1]. In contrast, IL-6 activities via the membrane bound IL-6 receptor, which is expressed by only a few cells in the body, are rather protective and regenerative [2]. Rose-John generated several Designer proteins, with which he analyzed the IL-6 trans-signaling signaling pathway. Hyper-IL-6 is a fusion protein, in which IL-6 and the soluble IL-6 receptor are covalently joined [3]. The sgp130Fc consists of the extracellular portion of the receptor subunit gp130 with the constant portion of a human IgG1 antibody. This protein inhibits specifically the IL-6 trans-signaling pathway without affecting the activities of IL-6 acting via the membrane bound IL-6 receptor [4]. The sgp130Fc protein was further developed to a therapeutic protein and under the name Olamkicept has successfully completed phase II clinical trials in patients with inflammatory bowel diseases [5] [6]. Currently, phase III clinical trials are planned.
