Garrett A. Perchetti is an American research scientist at the University of Washington School of Medicine, Virology Division in Seattle, WA. Perchetti's research focuses on respiratory viruses and methods to broaden access to COVID-19 testing[1][2][3]. He is an editor with the Journal of Medical Virology and does research with Next Generation Sequencing, Virology, and Infectious Diseases. [4]. Biotech companies like Sigma-Aldrich and Zymo Research have also utilized Perchetti's work on COVID-19 specimen pooling and assay performance to develop commercial laboratory diagnostic products[5][6]. In particular, his research regarding rapid antigen test sensitivity[7][8], especially on new COVID-19 variants,[9][10] has been critical to opening up schools and characterizes the limitations and utility of widespread, decentralized, rapid testing[11]. He also described the first instance of SARS-CoV-2 variants affecting rapid antigen test performance[12]
History[edit]
During global supply chain shortages in 2020, Perchetti demonstrated that transport and storage of SARS-CoV-2 in a much more available and affordable alternative (phosphate-buffered saline) was comparable to viral transport medium, relieving pressure on high-demand vital reagents involved in diagnostic testing[13]. In collaboration with Trevor Bedford[14], Helen Y. Chu[15], Keith R. Jerome[16], and Alexander L. Greninger [17][18], Perchetti detected the first instance of a COVID variant of concern in Washington State,( B.1.1.7-alpha) originally from the U.K.[19][20]. In efforts to address the global bottleneck of diagnostic reagents, Perchetti and colleagues designed an extraction-free method for detecting SARS-CoV-2 that dramatically reduced crucial reagent use, assay cost, and sample-to-answer turn around time[21][22]. The assay gain significant attention on the Cold Spring Harbor preprint server as one of its most downloaded publications, ranking in the top 0.08% of all published research outlets[23].
His research on Herpes Simplex Virus (HSV) has demonstrated for the first time that two species of HSV (oral and genital) can recombine to form new strains to increase the virus's genetic diversity[24][25]. Recently, his work has described SARS-CoV-2 infectivity demonstrating no culturable virus is detected beyond an RT-PCR cycle threshold of 31[26]. In collaboration with the Fred Hutchinson Cancer Research Institute, Perchetti reported on the incidence of viral drug resistance for the novel therapeutic Letermovir, designed and produced by Merck to treat CMV prophylactically in transplant patients[27]. In 2020, he teamed up with researchers from Johns Hopkins, the Milken Institute, and Seattle Children's Hospital and sequenced the first complete HMPV glycoproteins from Nepalese infants in efforts to increase the diversity of publicly available sequences of respiratory viruses[28]. Reuters featured Perchetti as a frontline healthcare worker during the COVID pandemic and in 2021 was nominated for Forbes 30 under 30[29]. Perchetti serves on microbiology editorial boards, is a referee in multiple medical journals for viral research, and is a member of AAAS, AMA, ASM, ACP and ASV. He attended Harvard University and the University of Washington receiving a BA (Art, Art History, and Design) and a BS (Microbiology),[30] and sits on the Eastlake Community Council in Seattle, WA[31]
