Jayamini Chandrasiri was born on 28 January 1967 in Colombo, Sri Lanka. Her father Upali Chandrasiri was a former Insurance Agent and her mother Malani Chandralatha is a Housewife. She has one brother Varuna Chandrasiri and one sister Champika Chandrasiri.
Jayamini first attended Somaweera Chandrasiri Primary School, Piliyandala, and then attended Piliyandala Central College in Piliyandala, where she excelled in Netball, representing the school.

In 1991 Jayamini married Pujitha Basnayake, with whom she had two sons, in 1996 and 1999.
