Pravat Kumar Mandal is an Indian neuroscientist. Presently, he is the Professor, Scientist, and Director-in-Charge of National Brain Research Centre and Honorary Professor at the Florey Institute of Neuroscience and Mental Health, Melbourne, Australia. His major area of research involves studying neuropsychiatric and neurodegenerative disorders, such as Alzheimer's and Parkinson's disease, using multimodal brain imaging techniques.[1][2]

Academic Profile

In 1995, Mandal received his Ph.D. from IIT Madras in “Exchange interactions in some bimetallic systems: experimental and theoretical approach.” From March 2004 to Jun 2008, he worked as an assistant professor in Psychiatry in The University of Pittsburgh Medical school, and from Feb 2011 to May 2016, as an Associate Prof. (Adj) (Radiology) at Johns Hopkins School of Medicine, Maryland, USA. He is currently an Honorary professor at the Florey Institute of Neuroscience and Mental Health, Melbourne, Australia.[3][4]

Mandal joined NBRC in 2008 as a scientist and faculty member. Today, he serves as scientist VII and the Director-in-Charge of the institute. He has established an independent lab at NBRC called the NeuroImaging and NeuroSpectroscopy (NINS) laboratory wherein his research team focuses on developing novel neuroimaging protocols, tools, and databases using multimodal neuroimaging approach for studying neurodegenerative and neuropsychiatric disorders, including Alzheimer's disease.[5]

Research Profile

Mandal's research focuses on studying early neurochemical–functional biomarkers for neurodegenerative disorders, including Alzheimer's and Parkinson's disease. His work uses non-invasive neuroimaging approach to investigate brain alterations in healthy aging and neurodegenerative disorders and develop neuroimaging tools and databases for processing of neuroimaging data.

Clinical Research and Discoveries

Mandal's team discovered early diagnostic markers of AD, wherein they showed that cognitive decline in AD is directly correlated to glutathione (GSH) depletion in the hippocampus and frontal and cingulate cortical regions.[6][7][8][9][10] His work explains the involvement of GSH depletion in the pathophysiology of MCI, demonstrating that increased oxidative stress induces GSH depletion and pH alteration.[11] Using 31P MRS, Mandal's team discovered that left hippocampal pH in AD-affected brains becomes alkaline.[12][13] His team also discovered that GSH molecule is expressed in closed and extended forms in the brain and identified that the closed GSH is often depleted in the brains affected with AD.[14][15][16][17][18][19]

Translational Research and Innovation

SWADESH

In October 2021, Mandal's team launched project SWADESH, which is the first large-scale multimodal neuroimaging database designed specifically for Indian population with big-data architecture and analytics for various disease categories under one platform. The database focuses on certified neuroimaging, neurochemical, neuropsychological data and analytics that are made accessible to researchers for managing brain disorders. Researchers from around the world can use this data/information for better defining a disease based on the analysis of imaging-based biomarkers.[20][21]

SWADESH was inaugurated by Dr. Jitendra Singh, Union Minister of State (Independent Charge) for the Ministry of Science and Technology on 19th November 2021. Upon the launch, Dr Rajesh Gokhale, Secretary, Department said, “Databases like SWADESH should be useful in conducting multimodal brain studies to understand Alzheimer’s disease and several neurological disorders.”[22]

ANSH

In October 2020, Mandal developed a brain neurochemical–neuroimaging database to help recognize cellular–molecular processes leading to various neurodegenerative–neuropsychiatric diseases. ANSH includes neurochemical levels, such as GSH and gamma aminobutyric acid (GABA), and MRI-based data for AD, mild cognitive impairment, and healthy subjects.[23]

KALPANA

In March 2020, Mandal developed KALPANA, an integrative MATLAB-based package for visualization, preprocessing, and quantitation of MRS data obtained using several acquisition methods. This free-for-public-access package aids in the diagnosis of brain disorders prior to their occurrence using oxidative stress and pH level analysis.[24]

BRAHMA

In July 2020, Mandal and his research team developed a novel brain template called “BRAHMA” using the brain scans of healthy Indians. BRAHMA provides details about the brain structures and regions and helps analyze brain images of various population groups (specific to Indian population). It also provides several image contrasts, segmentation atlases, and tissue maps for multimodal data and image analysis.[25][26][27]

BHARAT

In February 2019, the research team developed a Hadoop-based big data framework that integrates neuroimaging test outcomes to identify early diagnostic biomarkers of AD. The framework consists mainly of machine learning and statistical modeling. The work was published in Frontiers in Neurology[28] and covered by several platforms.[29]

Awards & honours

Mandal has been awarded with “Global Artificial Intelligence Award” (Category: Lifetime Achievement Award) by All India Council for Robotics & Automation[30] and TATA Innovation Fellow, Department of Biotechnology, Govt of India.

Throughout his quest, Mandal has delivered several talks and lectures at national and international level, including keynote addresses at IIT Indore (Topic: Digital Diagnosis using Neuroimaging Modalities: Tools and Applications; Dec 2017) as well as Anesthesia and Peri-Operative Care in the Seventh Age Anesthesia Association Annual Scientific Meeting, Stratford-upon-Avon, England, UK (Topic: Anesthetics simulating molecular mechanism of Alzheimer's disease; May 2013). Editorial Responsibilities

Mandal is the Regional Editor and Associate Editor for the Journal of Alzheimer's Disease[31]. In 2019, he was a Research Topic Editor for Frontiers in Neurology[32]. In his profession, he was also elected to be the Guest Editor for Journal of Alzheimer's Disease in 2010[33] and 2012[34] as well as for American Journal of Neuroradiology in 2014[35] Mandal is a member of several professional societies, including International Society for Magnetic Resonance in Medicine (Apr 2011–Jun 2013)[36], Society of Biological Psychiatry (May 2005–till date), and Society for Neuroscience (Jan 2003–till date).
